Proposal tailored to the purpose of analysis and the target object! GC/MS analysis of derivatized samples.

This document introduces a pretreatment method to enable the analysis of substances that are difficult to analyze using GC/MS. The pretreatment that converts difficult-to-analyze substances into analyzable substances is called "derivatization." Methods such as esterification, acylation, and silylation are used selectively based on the type and characteristics of the difficult-to-analyze substances. GC/MS measurements were conducted on citric acid solutions and citric acid solutions after esterification. It was found that citric acid, being a carboxylic acid, could not be effectively detected without derivatization, but after esterification, trimethyl citrate was detected, proving the presence of citric acid. By performing appropriate derivatization, more accurate analysis becomes possible. Undifferentiated mesenchymal stem cells have been revived!! A culture method that holds promise for application in bone augmentation therapy.

High-concentration purified mesenchymal stem cells have the ability to differentiate not only into mesenchymal cells but also into cells derived from neural crest stem cells. However, when cultured in a flat, adherent manner on a culture dish, there was a challenge of reduced proliferation ability and easy loss of undifferentiated characteristics as stem cells. The present invention has discovered that mesenchymal stem cells (which have lost their undifferentiated state) can form cell aggregates through suspension culture, maintaining (recovering) their proliferation ability and undifferentiated characteristics, and that these cell aggregates can be utilized as a source of stem cells.

Re-crosslinking of S-S bases is the key! A novel biotin labeling agent that stabilizes the structure and function of proteins.

Conventional biotin labeling randomly binds to functional groups of proteins, making it difficult to control the number of biotins bound and their binding positions. Additionally, the cleaved S-S bonds for labeling do not re-bridge, which poses a challenge as it can lead to changes in the higher-order structure of the protein and a decrease in activity. In this technology, biotin is introduced to the Br group of dibromo-pyridazine dione, and the re-bridging of S-S bonds occurs simultaneously with labeling. This stabilizes the higher-order structure of the protein and preserves its activity. The number of bound biotins can be selected as either one or two, and it can be used for controlling the orientation of proteins by utilizing the binding characteristics with streptavidin.

Fluorescent dyes that can be widely used from biological systems to metals.

Triptanslin is known as an antibacterial agent extracted from the plant indigo. The inventors of this invention chemically synthesized triptanslin derivatives that cannot be obtained from natural sources and were investigating the structure-activity relationship regarding antibacterial properties when they discovered that introducing an amino group at the 2-position of triptanslin resulted in strong fluorescence. This invention relates to fluorescent reagents of triptanslin. **Characteristics of Triptanslin Derivatives** - 2-Aminotryptanslin (T2NH2) can be excited in the visible light wavelength range with minimal damage to cells and changes its fluorescence color in response to the surrounding environment (polarity). Additionally, through fluorescence resonance energy transfer (FRET) with pyrene, red fluorescence derived from T2NH2 is observed when FRET-on occurs, but in the presence of certain metal ions such as mercury, it becomes FRET-off and changes to blue fluorescence derived from pyrene. - 2-Hydroxytriptanslin emits light in the wavelength range of the biological window (650-900 nm), which has excellent biocompatibility with minimal influence from proton dissociation in the excited state and absorption by water or biological substances.
